I am planning to tow my 1927 Coach on a tow dolly for a round trip of about 50 miles. The front wheels would be secured to the dolly and the transmission would be in neutral. I wouldn't exceed 45 mph. What concerns, if any, should I have?

The first thing that would concern is is going that fast. I would not exceed 30 or 35 MPH....and less if the car has not been driven on the road for years.

I suggest erring on the safe side and rent a full trailer for the trip. Cheap insurance against some mishap.

Making turns with the tow dolly and the car having a very wide and longer front fenders could be difficult.
