Does anyone know if a manifold (intake/exhaust combination)for a 216 is different from a 235? Are they interchangeable?

That is correct. The carburetor hole is larger, as well as the spacing between its mounting bolts, also.

I'm not sure if you can use a 216 manifold on a 235 or not, given that the port holes are a different size.

I'm not 100 percent on this but I believe it to be correct. As soon as some of those more knowledgeable finally roll outta bed, you'll get a better answer. And, we'll know if I got it right or not. Agrin

Charlie is correct, you can use a 216 intake/exhaust manifold on a 235. You will need to use a 216 carb for the reasons already mentioned. Most of the vendors sell alignment rings that have the 235 size on one side and the 216 size on the other.

O.K. now everyone in the world is going to know how ignorant I am. What are port holes? Exhaust and intake openings on the engine block???

By that question you have a lot to learn and this forum is the place to start.
The inlet and exhaust ports are in the head and are the passages that connect the manifold openings to the combustion chamber. The valves close off the chamber end.

The second photo down shows a head with the valve layout. One exhaust and one intake per cyclinder. The intake and exhaust manifold bolts to the head.
